Il s\u2019agit d\u2019une mutation g\u00e9n\u00e9tique extr\u00eamement rare, responsable d\u2019une maladie musculaire touchant une centaine de personnes et qui cr\u00e9e une immunit\u00e9\u00a0\u00bb naturelle contre le VIH.<\/em><em> C\u2019est ce qu\u2019ont rapport\u00e9 jeudi 29 ao\u00fbt des chercheurs espagnols : ils esp\u00e8rent que cette avanc\u00e9e sera une piste pour de nouveaux m\u00e9dicaments anti-vih.<\/em>.<\/p>\n<p><strong>Une nouvelle mutation bien plus rare<\/strong><\/p>\n<p>La nouvelle mutation concerne un\u00a0 <span style=\"color: #ff0000;\">g\u00e8ne (Transportine-3 ou TNPO3)<\/span> beaucoup plus rare : elle a \u00e9t\u00e9 <span style=\"color: #ff0000;\">d\u00e9couverte il y des ann\u00e9es chez une m\u00eame famille en Espagne<\/span>, atteinte d&rsquo;une maladie musculaire ultra-rare, appel\u00e9e <span style=\"color: #0000ff;\">dystrophie musculaire des ceintures de type 1F.<\/span><\/p>\n<p>Les m\u00e9decins se sont aper\u00e7us que <span style=\"color: #0000ff;\">des chercheurs sur le VIH s&rsquo;int\u00e9ressaient s\u00e9par\u00e9ment au m\u00eame g\u00e8ne,<\/span> car il<span style=\"color: #0000ff;\"> joue un r\u00f4le dans le transport du virus \u00e0 l&rsquo;int\u00e9rieur des cellules<\/span>.Ils ont donc contact\u00e9 des g\u00e9n\u00e9ticiens de Madrid, qui ont eu l&rsquo;id\u00e9e de tenter d&rsquo;infecter, en laboratoire, du sang des membres de cette famille espagnole avec le virus du sida.<\/p>\n<p><strong>Des lymphocytes qui r\u00e9sistent au virus<\/strong><\/p>\n<p><span style=\"color: #ff0000;\">L&rsquo;exp\u00e9rience a livr\u00e9 une surprise<\/span> : les<span style=\"color: #ff0000;\"> lymphocytes<\/span> de ceux qui avaient cette maladie musculaire ultrarare \u00e9taient <span style=\"color: #ff0000;\">naturellement r\u00e9sistants au VIH. Le virus n&rsquo;arrivait pas \u00e0 y p\u00e9n\u00e9trer<\/span>.<\/p>\n<p>\u00ab\u00a0C<span style=\"color: #0000ff;\">ela nous aide \u00e0 comprendre beaucoup mieux le transport du virus dans la cellule<\/span>\u00ab\u00a0, explique\u00a0 \u00e0 l&rsquo;AFP <span style=\"color: #0000ff;\">Jos\u00e9 Alcami, le virologue de l&rsquo;Institut de sant\u00e9 Carlos III<\/span> \u00e0 Madrid qui a men\u00e9 ces recherches publi\u00e9es dans la revue am\u00e9ricaine\u00a0<a href=\"https:\/\/journals.plos.org\/plospathogens\/article?id=10.1371\/journal.ppat.1007958\">PLOS Pathogens<\/a>.<\/p>\n<p><strong>Des exceptions encore tr\u00e8s mal comprises<\/strong><\/p>\n<p><span style=\"color: #ff0000;\">Le VIH est certes le mieux connu de tous les virus<\/span>, dit-il, \u00ab\u00a0mais il y a encore<span style=\"color: #ff0000;\"> beaucoup de choses qu&rsquo;on conna\u00eet mal.<\/span> Par exemple, on ne sait pas<span style=\"color: #ff0000;\"> pourquoi 5% des patients qui sont infect\u00e9s ne d\u00e9veloppent pas de sida.<\/span> Il y a <span style=\"color: #0000ff;\">des m\u00e9canismes de r\u00e9sistance \u00e0 l&rsquo;infection qu&rsquo;on comprend tr\u00e8s mal\u00a0\u00bb.<\/span><\/p>\n<p><span style=\"color: #ff0000;\">Le chemin est encore long pour exploiter cette faille<\/span> afin de produire un nouveau m\u00e9dicament.
